STAB2 Gene encodes a large transmembrane receptor protein that may be involved in functions such as angiogenesis, lymphocyte homing, cell adhesion, or receptor scavenging. STAB2 is the primary clearance receptor for hyaluronic acid, and studies suggest that STAB2 could be a potential target for anti-tumor therapies. Blocking STAB2 function could prevent tumor metastasis by increasing hyaluronic acid levels in circulation [1]. This protein is primarily expressed in the sinusoidal endothelial cells of the liver, spleen, and lymph nodes.
The Stab2-iCre-EGFP mouse model is generated by knocking in a high-recombination activity iCre element (codon-optimized Cre) into the stop codon of the endogenous Stab2 gene in mice. The expression pattern of the iCre recombinase is similar to that of the endogenous Stab2 gene. Using 2A peptides (P2A and T2A) avoids the issues of reduced protein activity and downstream gene expression levels in multi-gene expression processes. Additionally, this strain carries a green fluorescent protein (EGFP) expression element, which can be used for lineage tracing of Stab2-positive cells. When Stab2-iCre-EGFP mice are crossed with mice containing loxP sites, Cre recombinase-mediated recombination between loxP sites is expected to occur in the offspring's STAB2-expressing cells.
