The NLRC5 gene encodes a protein known as the NOD-like receptor family CARD domain containing 5, which acts as the primary transcriptional activator for MHC class I genes. It is predominantly expressed in lymphoid tissues—specifically within T cells, B cells, and macrophages—where it shuttles into the nucleus to orchestrate the "CITA" (Class I Transactivator) complex. By binding to the promoter regions of MHC class I and related genes (like β2-microglobulin), NLRC5 ensures the cellular machinery is ready to present antigens to CD8+ T cells, making it a cornerstone of the adaptive immune response [1]. Dysregulation of NLRC5 is heavily associated with cancer evasion, where its downregulation allows tumors to "hide" from the immune system, as well as various inflammatory and autoimmune diseases resulting from improper immune activation.
Nlrc5-iCre mice were generated using gene editing technology to replace the TGA stop codon with the P2A-iCre cassette. Under the control of the Nlrc5 gene's regulatory elements, the iCre recombinase (a codon-optimized Cre recombinase) is expressed. When Nlrc5-iCre mice are crossed with mice containing loxP sites, Cre recombinase-mediated recombination between loxP sites is expected to occur in the lymphoid tissues expressing the NLRC5 transcription factor, particularly in T cells, B cells, and macrophages in the offspring.
