The CALCA gene, expressed in various tissues including the thyroid and parathyroid glands as well as sensory neurons, undergoes tissue-specific alternative RNA splicing to produce the peptide hormones calcitonin, calcitonin gene-related peptide (CGRP), and katacalcin. Calcitonin functions in calcium regulation and phosphorus metabolism, while CGRP acts as a vasodilator and antimicrobial peptide, and katacalcin also plays a role in lowering calcium levels. The CALCA gene has been associated with diseases such as hepatocellular carcinoma and reflex sympathetic dystrophy, and CGRP is notably involved in conditions like migraine.
Calca-iCre mice were generated using gene editing technology to integrate a Kozak-iCre-P2A expression cassette into the upstream of ATG start codon of the mouse endogenous Calca gene. Under the control of the Calca gene's regulatory elements, the iCre recombinase (a codon-optimized Cre recombinase) is expressed. When Calca-iCre mice are crossed with mice containing loxP sites, Cre recombinase-mediated recombination between loxP sites is expected to occur in the neurons expressing calcitonin protein in the offspring.
