The collagen type II alpha 1 chain (COL2A1) gene encodes the alpha-1 chain of collagen type II, a fibrillar collagen found in cartilage and the eye's vitreous humor. Collagen type II is closely related to the structure and strength of connective tissue and is found primarily in cartilage, vitreous humor, inner ear, and the gelatinous material (nucleus pulposus) in the central part of the intervertebral disc between the vertebrae of the spine.
This strain was generated by integrating the mouse Col2a1 gene promoter-regulated codon-improved Cre recombinase (iCre) gene expression cassette into the mouse genome. This results in the conditional expression of iCre in chondrocytes, under the control of the Col2a1 promoter. When bred with mice containing a loxP site-flanked sequence, Cre recombinase-mediated deletion of the flanked sequence will occur specifically in chondrocytes in the offspring. Both homozygous and heterozygous H11-Col2a1-iCre mice are viable and fertile.

検証 Data
1. Method
H11-Col2a1-iCre mice were crossed with Rosa26-LSL-tdTomato mice, which conditionally express tdTomato, to generate double-heterozygous offspring. In the offspring, Cre recombinase-mediated deletion of the expression termination element (LSL) resulted in tdTomato protein expression in Cre-positive cells. Ankle, knee, and elbow joint tissues from 7-week-old offspring mice were collected and examined for tdTomato protein expression by frozen sections and immunohistochemistry to determine Cre recombinase activity.

3. Result
(1)Expression of Cre recombinase in ankle joints
Abundant orange spontaneous fluorescence signals were observed in the ankle joints of Cre+ mice, indicating strong Cre recombinase activity in the ankle joints of this model. No tdTomato protein signals were detected in the control group (Cre-), suggesting no expression of Cre recombinase.

(2)Expression of Cre recombinase in elbow joints
Abundant orange spontaneous fluorescence signals were observed in the elbow joints of Cre+ mice, indicating strong Cre recombinase activity in the elbow joints of this model. No tdTomato protein signals were detected in the control group (Cre-), suggesting no expression of Cre recombinase.

(3)Immunohistochemistry of knee joints
Results showed that corresponding cells in the knee joints of Cre+ mice exhibited strong tdTomato-positive signals.

4. Summary
In H11-Col2a1-iCre mice, Cre recombinase is primarily expressed in chondrocytes in joints such as the ankle, knee, and elbow. Internal data show that Cre recombinase is also expressed in small amounts in other tissues such as the heart, lungs, tail, brain, brown fat, and sternum (not shown in this manual). In conclusion, this model is a specific Cre mouse that primarily targets chondrocytes, but it is important to note that leakage to other tissues may potentially affect the actual results when using this model.
