NKG mice (Catalog number: C001316) are severely immunodeficient mice generated by Cyagen through deleting the Il2rg gene from NOD-Scid mice. NKG mice exhibit deficiency of mature T cells, B cells, and functional NK cells, reduced complement activity, and weak phagocytosis of human-derived cells by macrophages, which are well suited for the transplantation of human hematopoietic stem cells (HSC), peripheral blood mononuclear cells (PBMC), adult stem cells and tissues, and patient-derived xenografts (PDX).
huHSC-NKG mice are immune system humanized mouse models constructed by transplanting human hematopoietic stem cells (HSC) into NKG mice after sub-lethal dose irradiation. This model has a long lifespan and can successfully reconstitute various immune cells. In addition, huHSC-NKG mice typically do not develop graft-versus-host disease (GvHD) and the reconstitution period can be long.
The huHSC-NKG-ProM mouse is the latest enhanced version of the huHSC-NKG series, where "Pro" indicates the enhanced series and "M" specifically refers to myeloid cells. This mouse utilizes advanced neonate technology and has been process-optimized to allow for the development of lymphoid T cells and B cells, as well as myeloid cells such as monocytes. It is suitable for research on immune checkpoint inhibitors and drugs that target myeloid cells.

検証 Data
1. Survival and Growth Curves
During the process of human immune system reconstitution, the body weight of NKG mice was maintained at 18-22 grams, and the survival rate at 18 weeks post-reconstitution was 85%.

2. Reconstitution of Human Leukocytes in Peripheral Blood
Transplantation of human umbilical cord blood-derived hematopoietic stem cells (HSCs) into NKG mice for human immune system reconstitution resulted in a stable reconstitution efficacy, with the proportion of human-derived CD45+ leukocytes reaching approximately 40%-60%.

3. Reconstitution of Human T Cells and B Cells in Peripheral Blood
The gradual reconstruction of T cells commenced from the 10th week post-transplantation of human hematopoietic stem cells (HSCs), with the mean T cell content reaching 40% by the 16th week. The proportion of B cells ranged from approximately 60-80% during weeks 6-8 post-reconstruction. Subsequently, as the T cell proportion increased, the B cell proportion gradually decreased to 40-60%.

4. Reconstitution of Human NK Cells and Monocytes in Peripheral Blood
The average reconstitution proportion of Natural Killer (NK) cells was below 5%, while monocytes in peripheral blood achieved a reconstitution proportion of 5-10%. Given that macrophages are primarily derived from monocytes, monocyte detection is therefore the main focus in peripheral blood samples.
