This mouse strain is a fluorescent reporter mouse carrying a red fluorescent protein (tdTomato) gene expression cassette, with tdTomato protein expression regulated by the CAG promoter. The CAG promoter is a modified composite promoter consisting of the chicken β-actin gene promoter, the cytomegalovirus (CMV) early enhancer, the rabbit β-actin gene splice acceptor, and the bovine globulin polyA signal sequence. Renowned for driving robust and stable gene expression in multiple mammalian cell types, this promoter induces consistent tdTomato expression across systemic tissues and organs.
RCL-tdTomato mice are generated via genome editing to knock the CAG-LSL-tdTomato cassette into the ROSA26 safe harbor locus. Upstream of the tdTomato gene lies a loxP-Stop-loxP (LSL) cassette, which blocks the transcription and expression of tdTomato. Upon Cre recombination, cells expressing Cre recombinase (and their future cell lineages) exhibit strong tdTomato fluorescence. This model is suitable for validating Cre mice with high recombination efficiency and potential expression leakage, as well as for other cell lineage tracing applications.

検証 Data
1. Method
Crossing RCL-tdTomato mice (conditionally expressing tdTomato fluorescent protein) with Cd19-iCre mice (B lymphoid lineage-specific Cre mice) generates double heterozygous offspring. Cre recombinase-mediated deletion of the expression termination cassette (LSL) drives tdTomato protein expression in Cre-positive cells. At 5 weeks of age, peripheral blood, spleen, and bone marrow tissues from offspring mice were collected, and tdTomato expression in T and B cells was detected by fluorescence-activated cell sorting (FACS) to confirm Cre recombinase activity.

(1)Cre recombinase expression in peripheral blood B lymphocytes, T lymphocytes

Weak Cre recombination was detected in splenic T lymphocytes of Cre+ mice, potentially linked to nonspecific Cre recombinase expression or expression leakage. Internal data showed that RCL-tdTomato[CKI/+];Cd19-iCre[KI/+] mice exhibited lower recombination efficiency in splenic T lymphocytes compared to Rosa26-LSL-tdTomato[CKI/+];Cd19-iCre[KI/+] mice. Given that Cd19-iCre mice are B lymphocyte-specific Cre mice, these results further indicate that RCL-tdTomato mice may be more suitable than Rosa26-LSL-tdTomato mice (Product No.: C001476) for detecting Cre mice with high recombination efficiency and potential expression leakage.
(3)Cre recombinase expression in bone marrow B lymphocytes

4. Summary
In RCL-tdTomato mice, tdTomato fluorescence can be detected in Cre-positive cells after Cre recombination. This model is suitable for validating Cre mice with high recombination efficiency and prone to expression leakage.
