huIL15RA Mouse

The IL15RA gene, located on chromosome 10, encodes the interleukin-15 receptor subunit alpha, a high-affinity binding protein essential for mediating the biological activities of IL-15. This gene is widely expressed across various tissues, including lymphoid organs (thymus and spleen), the gastrointestinal tract, skeletal muscle, and liver, as well as by specific cell types such as monocytes, dendritic cells, and activated T and B cells. The encoded protein is unique for its "cis" and "trans-presentation" mechanisms, where it binds IL-15 within the endoplasmic reticulum and transports it to the cell surface to signal to neighboring cells expressing IL-2/IL-15 receptor β- and γ-chains. This function is critical for the proliferation and survival of Natural Killer (NK) cells and memory CD8⁺ T cells, thereby regulating innate and adaptive immune homeostasis ^[1]. Pathologically, dysregulation of IL15RA is associated with various autoimmune diseases like rheumatoid arthritis (RA) and psoriasis, as well as inflammatory bowel diseases (IBD) and several hematological malignancies ^[2].

The huIL15RA mouse is a humanized model constructed through gene-editing technology, in which the mouse Il15ra endogenous extracellular domain is replaced with the human IL15RA extracellular domain. This model can be used for research into the pathological mechanisms and related therapeutic approaches for autoimmune diseases, inflammatory diseases, and various hematologic malignancies, as well as for screening, development, and preclinical evaluation of IL15RA-targeted drugs.
