Crhbp, short for corticotropin-releasing hormone-binding protein, plays a crucial role in regulating corticotropin release. It is a key regulator in the neuroendocrine, autonomic, and stress adaptation processes, functioning as a negative feedback regulator within the hypothalamus-pituitary-adrenal (HPA) axis by binding CRH with high affinity to inhibit CRH-induced ACTH secretion from pituitary cells [4].
In tumor-related research, in most tumors, Crhbp is downregulated and is identified as an important factor for predicting the prognosis of cancer patients. Overexpression of Crhbp significantly inhibits cell proliferation of LUAD, LIHC, and KIRC cell lines, and also reduces cell mobility in KIRC and HCC cells, suggesting its anti-tumorigenic properties [1]. In HCC, Crhbp downregulation may be due to p62-mediated autophagy, and Crhbp exerts anti-HCC effects by reducing cyclin B2 expression and dissociating cyclin B2-CDK1 complex [2]. In ccRCC, overexpression of Crhbp suppresses cell proliferation, migration, invasion, and apoptosis, and it may be an effective target for treating ccRCC patients [3].
In conclusion, Crhbp is essential for the normal regulation of the HPA axis. Its dysregulation is closely associated with tumor development, and its downregulation in various tumors is related to poor prognosis. Research on Crhbp provides potential targets for cancer treatment, helping to better understand the mechanisms of tumorigenesis and develop new treatment strategies.
