Il1b, encoding interleukin-1β, is a key cytokine in the inflammatory response pathway. It is crucial for the host-response to pathogens, but also contributes to damage in chronic diseases and acute tissue injury. IL-1β does not follow the conventional ER-Golgi secretion route, and its release mechanism is still being explored [2].
Single-cell profiling has identified IL1Bhi macrophages associated with inflammation in PD-1 inhibitor-induced inflammatory arthritis. These macrophages have enhanced NLRP3 inflammasome activity and communicate with exhausted CD8+ T cells, suggesting a different cellular and molecular pathway compared to rheumatoid arthritis and highlighting IL1Bhi macrophages as a potential therapeutic target [1]. In addition, certain IL1B gene polymorphisms are associated with diseases such as keratoconus, essential tremor, and may play a role in miscarriage due to BaP/BPDE exposure [3,4,5].
In conclusion, Il1b is essential for the inflammatory response, and its study through genetic models, especially in relation to various diseases like PD-1 inhibitor-induced arthritis, keratoconus, essential tremor, and miscarriage, has provided insights into disease mechanisms. Understanding Il1b function can potentially lead to new therapeutic strategies for these conditions.
