Marchf2, also known as membrane associated ring-CH-type finger 2, is a gene that has been implicated in multiple biological processes. It is a member of the MARCH protein family of RING-CH finger E3 ubiquitin ligases, which play important roles in regulating the levels of proteins on the cell surface [2,3,6]. This family of proteins is involved in processes such as protein trafficking, inflammation response, and antiviral immunity [5,7].
In the context of diseases, a panel of eight exLRs including Marchf2 was developed as a diagnostic signature for pancreatic ductal adenocarcinoma (PDAC), showing high accuracy in detecting the disease, even at resectable stage I/II [1]. Marchf2 has also been associated with glucose metabolism-six single nucleotide polymorphisms (SNPs) in genes including Marchf2 reached genome-wide significance in relation to 2-hour postprandial proinsulin levels, suggesting its potential as a biomarker for prediabetes prevention [8]. In addition, it has antiviral functions: human March2 (a product of the Marchf2 gene) restricts HIV-1 infection in primary CD4+ T cells and can prevent cell-to-cell transmission of HIV-1, and it along with March1 and March8, blocks Ebola virus envelope glycoprotein cleavage [2,3]. In triple-negative breast cancer, March2 acts as a SNAIL E3 ligase, suppressing metastases by promoting SNAIL degradation [4]. In hepatocellular carcinoma, transcription factor KLF15 activates March2, which then boosts cell proliferation and epithelial-mesenchymal transition, presenting diagnostic significance [5].
In conclusion, Marchf2 plays diverse and significant roles in various biological processes and disease conditions. Its involvement in PDAC diagnosis, glucose metabolism-related biomarker potential, antiviral functions, and cancer-related processes such as metastasis and cell proliferation regulation highlights its importance. The study of Marchf2 through various research models, though not specifically detailed here in terms of KO/CKO mouse models, helps in understanding disease mechanisms and could potentially lead to new therapeutic strategies.
