Vsig1, a cell-cell adhesion molecule, is a member of the immunoglobulin superfamily. It is preferentially expressed in the stomach, testis, and certain cancers such as gastric, esophageal, and ovarian cancers [4]. In the stomach, it is involved in the proper differentiation of glandular gastric epithelia, being restricted to the adherens junction of the glandular epithelium [4]. In testis, it is located on the testicular germ cell surface and its extracellular domain interacts with Sertoli cell membrane protein while its PDZ-binding motif forms a complex with ZO-1 [7].
In gastric cancer, several studies have explored its role. Loss of VSIG1 expression is significantly correlated with poor overall and disease-free survival, and negative VSIG1 expression is related to tumor size, T, and M stage, suggesting it may be a prognostic marker and potential therapeutic target [5]. Also, its membrane-cytoplasmic translocation can indicate epithelial-mesenchymal transition due to the activation of the canonical Wnt/β-catenin signaling pathway [1]. In hepatocellular carcinoma, VSIG1 may be involved in the lineage shift to gastric-type HCC, and the E-cadherin/VSIG1 complex may help suppress tumor growth [6]. In colon cancer, VSIG1 restricts the development of pro-metastatic states, in part by inhibiting YAP/TAZ-TEAD signaling [2]. A study on VSIG1 knockout (KO) mice found that VSIG1 may not play essential roles in spermatogenesis and normal testis development, function, and maintenance in mice [3].
In conclusion, Vsig1 has important functions in the proper differentiation of glandular gastric epithelia and in testis cell interactions. In cancer, its expression and translocation are associated with disease progression in gastric and hepatocellular carcinomas, and it has an anti-metastatic role in colon cancer. The Vsig1 KO mouse model has been valuable in revealing its dispensability in spermatogenesis and male fertility in mice, contributing to our understanding of its role in specific biological processes and disease conditions.
