Slc24a3, belonging to the solute carrier family 24, is a sodium-calcium regulator of cells [6,8]. It may be involved in maintaining the balance of intracellular calcium ions, a process closely related to many cellular functions such as cell contraction, signal transduction, and gene expression regulation [1,7].
Genetic studies have associated Slc24a3 with multiple diseases. In fibromuscular dysplasia, a transcriptome-wide association analysis in arteries identified it as an associated locus [1]. In migraine-first patients, it was among the key risk loci, and pathway analyses highlighted retinol metabolism and steroid hormone biosynthesis as important pathways [2]. In triple-negative breast cancer, a 10-gene signature including Slc24a3 was identified to predict invasive disease-free survival [3]. It was also part of a six-gene prognostic signature in cervical cancer, and closely associated with the immune response and tumour immune microenvironment [4]. In acute myeloid leukemia, a 5-gene prognostic signature containing Slc24a3 was developed to predict prognosis and decipher the immune microenvironment [5]. In gynecologic cancer, its down-regulation in tumor tissues was associated with patient survival rates [6]. In colon adenocarcinoma, a prognostic model based on calcium extrusion-related genes including Slc24a3 was constructed, with high risk scores related to poor prognosis [7]. In cervical squamous cell carcinoma and endocervical adenocarcinoma, low SLC24A3 expression was associated with higher overall survival [8].
In conclusion, Slc24a3 is a crucial gene involved in cell calcium regulation. Its abnormal expression is associated with various diseases, including fibromuscular dysplasia, migraine, breast cancer, cervical cancer, acute myeloid leukemia, gynecologic cancer, and colon adenocarcinoma. These disease-related associations highlight the importance of studying Slc24a3 to understand disease mechanisms and potentially develop new diagnostic and therapeutic strategies.
