Bmp4, short for Bone Morphogenetic Protein 4, is a crucial protein in the BMP signaling pathway. This pathway is involved in multiple biological processes, including early embryonic morphogenesis, especially in the epiblast, as well as the development of extra-embryonic lineages such as trophectoderm [2,3].
In corneal neovascularization (CNV), Bmp4 inhibits the formation of tip cells by reducing the generation of neutrophil extracellular traps (NETs) through the NADPH oxidase-2 (NOX-2) pathway, disrupting the tip-stalk cell balance and thus inhibiting CNV, suggesting it as a potential therapeutic target [1]. In trophoblast development, BMP4-based protocols can convert pluripotent stem cells into trophoblast in vitro, and understanding its role helps model placental development [2,3]. Regarding non-syndromic cleft lip with or without cleft palate (NSCLP), a meta-analysis showed that BMP4 rs17563 polymorphism may be a risk factor in Chinese and Brazilian populations [4]. BMP4 also enhances cell migration and promotes epithelial-mesenchymal transition (EMT) through the Smad1 signaling pathway, which is associated with NSCLP [7]. In breast cancer, BMP4 suppresses metastasis by inhibiting cholesterol biosynthesis, and it may be a predictive biomarker for statin therapy benefit [5]. In endometrial stromal cells, BMP4 affects decidualization by regulating FOXO1 expression, and its down-regulation may contribute to recurrent spontaneous abortion (RSA) [6]. In glioblastoma, BMP4 inhibits invasion by promoting E-cadherin and claudin expression through SMAD signaling [8]. In human preimplantation embryos, BMP4 supplementation triggers apoptosis [9].
In conclusion, Bmp4 is essential in various biological processes such as embryonic development, cell differentiation, and apoptosis. Its role in diseases like CNV, NSCLP, breast cancer, RSA, and glioblastoma has been revealed through different research models. Understanding Bmp4 not only deepens our knowledge of normal biological functions but also provides potential therapeutic directions for these diseases.
