遺伝子研究の概要
Lrrc37a is a gene located within the 17q21.31 locus. It is a membrane-associated protein that has been implicated in multiple biological functions. In terms of associated pathways, it may be involved in cellular migration, chemotaxis, and astroglial inflammation [2]. It also seems to play a role in the context of neurodegenerative diseases, making it of significant biological importance. Genetic models such as gene knockout (KO) or conditional knockout (CKO) mouse models could potentially be valuable for further studying its functions.
In post-traumatic stress disorder (PTSD) and major depressive disorder (MDD), Lrrc37a was among the genes identified through single-nucleus RNA-seq analysis of the dorsolateral prefrontal cortex. In MDD samples, it was one of the differentially expressed genes, while in PTSD, genes from the 17q21.31 locus including Lrrc37a2 were identified, highlighting cell-type-specific mechanisms of brain stress response [1]. In Parkinson's disease (PD), the 17q21.31 locus is genetically associated with the disease. Protective sub-haplotypes were associated with increased Lrrc37a/2 copy number and expression in human brain tissue. Lrrc37a/2 was found to interact directly with soluble α-synuclein and co-localize with Lewy bodies in PD brain tissue, indicating its contribution to the association between the 17q21.31 locus and PD [2]. In progressive supranuclear palsy (PSP), γ duplication within the 17q21.31 region up-regulates the expression of Lrrc37a/Lrrc37a2 in neuronal cells, and the copy number of γ is associated with the risk of PSP [3].
In conclusion, Lrrc37a is a gene with functions related to cellular processes like migration and inflammation, and is associated with neurodegenerative diseases such as PD and PSP, as well as mental disorders like PTSD and MDD. Studies on Lrrc37a using KO or CKO mouse models could potentially further clarify its role in these disease conditions, providing insights into the underlying biological mechanisms and potentially leading to new therapeutic strategies [1,2,3].
