Tspan5, a member of the tetraspanin family, is involved in multiple biological functions. It can interact with specific partner proteins in tetraspanin-enriched microdomains, regulating their subcellular localization and function. It is associated with pathways such as Notch signaling, and plays important roles in processes like cell fusion, epithelial-mesenchymal transition (EMT), and synaptic function, with implications for various diseases [1,2,4].
In placental trophoblastic cells, Tspan5 promotes the EMT process to regulate syncytialization by activating Notch signalling. Knockdown of Tspan5 inhibits cell fusion and EMT-related protein levels [1].
In hepatocellular carcinoma (HCC), Tspan5 promotes EMT and tumour metastasis by activating Notch signalling, and its high expression is associated with poor prognosis. Lentivirus-mediated alteration of Tspan5 expression in HCC cells shows its role in promoting cell migration and tumour metastasis in vitro and in vivo [2].
In alcohol-related disorders, ethanol and acamprosate treatment can down-regulate Tspan5 expression, influencing serotonin and kynurenine concentrations [3].
In the nervous system, Tspan5 knockdown in mouse pyramidal excitatory neurons causes a decrease in spine number due to spine maturation defects, and it promotes neuroligin-1 surface clustering [4]. In rat hippocampal neurons, Tspan5 promotes AMPA receptor exocytosis by interacting with the AP4 complex [5].
In gastric cancer, Tspan5 is down-regulated, and its up-regulation inhibits cell proliferation and tumour growth by modulating cell cycle-related proteins [6].
In HCC, depletion of Tspan5 induces oncogene-induced senescence through the p16INK4a/pRb pathways and affects actin polymerization and MRTF-A-FLNA complex formation [7].
In conclusion, Tspan5 is crucial in multiple biological processes and diseases. Its role in cell fusion, EMT, synaptic function, and tumour-related processes has been revealed through various functional studies including knockdown experiments. These findings contribute to understanding normal development and disease mechanisms, such as in placental development, cancer metastasis, and alcohol-related disorders, providing potential therapeutic targets.
